Overall-survival findings are promising but require scrutiny of the prespecified testing plan.

SANOVO randomized 326 patients, 162 to the combination and 164 to placebo plus osimertinib. In the IHC 2+/3+ intention-to-treat group, blinded-review median PFS was 24.9 versus 13.8 months (HR 0.56, 95% CI 0.43–0.74; P<0.0001). In the high-MET subset, median PFS was 24.8 versus 11.1 months (HR 0.45, 95% CI 0.31–0.66). The reported OS HR was 0.48 (95% CI 0.33–0.68), with medians 44.5 and 34.9 months; 49 versus 83 deaths had occurred. Adverse-event discontinuation on combination was 14.8%; fatal adverse events excluding progression were 4.3% versus 4.8%. The PFS cutoff was 31 July 2026. This disclosure adds quantitative data to the August top-line result.

Deep Analysis

Randomization against active first-line osimertinib and biomarker selection make the PFS signal compelling within the Chinese trial. A roughly 11-month absolute median PFS gain in the overall selected group is materially larger than many add-on kinase strategies. Yet the high-MET subset and broader IHC 2+/3+ group must not be conflated. The release does not establish how OS multiplicity was controlled, nor does it provide full grade-specific toxicity, dose-intensity or post-progression treatment tables. First-line MET overexpression may reflect intrinsic bypass biology, but assay cutoffs, sample timing and geographic prevalence determine who would qualify elsewhere. SANOVO is domestic-only evidence. It complements the global post-osimertinib SAFFRON trial, reported separately on 25 October, but does not establish the same effect in a global first-line population. HUTCHMED plans a China supplemental application; filing is not approval.

Company and Product Background

HUTCHMED discovered savolitinib, an oral MET kinase inhibitor, and develops it with AstraZeneca. Osimertinib inhibits mutant EGFR. MET overexpression can provide bypass signaling in EGFR-mutant non-small-cell lung cancer. The study enrolled EGFR exon-19 deletion or L858R disease with MET expression defined by immunohistochemistry.

Signal Extraction

Interpretation A: concurrent MET inhibition prevents or delays EGFR bypass resistance; randomized PFS and OS point estimates support it.

Interpretation B: benefit is concentrated in a stringent high-MET subset or influenced by care after progression; incomplete assay and downstream-treatment detail leaves this open.

Upgrade: full statistical hierarchy, mature OS, safety and quality-of-life tables, centrally reproducible IHC, and multinational validation. Downgrade: smaller benefit outside China or cumulative hepatic and edema toxicity. Falsification: a biomarker-matched global trial finds no clinically meaningful benefit.

InSilens Take

Inference: SANOVO makes first-line EGFR–MET dual blockade a strong China-specific precision-oncology candidate. The size of the OS signal should remain provisional until its testing framework and full dataset are public.

Signal Assessment

Signal Importance: 5 of 5. Signal Direction: positive for PFS; OS uncertain. Confidence in Facts: high. Confidence in Interpretation: moderate.

New Phase III data presented on 26 October show a large progression-free survival benefit from savolitinib plus osimertinib over osimertinib alone in untreated EGFR-mutant, MET-overexpressing advanced NSCLC in China.
