Four weeks of oral NEK7-directed molecular glue MRT-8102 produced 80\u201390% median NEK7 degradation and broad reductions in inflammatory biomarkers. Human target engagement is clear from the company disclosure; plaque improvement or fewer cardiovascular events has not been shown.<\/p>\n<p>Participants received 5, 20 or 40 mg daily or placebo for four weeks, followed by four weeks of safety observation. Company-reported median reductions across treated groups included calprotectin 56%, S100A12 46%, serum amyloid A 51%, IL-6 54%, hsCRP 85% and lipoprotein(a) 24%. Serious adverse events were not reported; treatment-emergent adverse events occurred in 33% of active versus 30% of placebo recipients. Arm-level denominators, adjusted between-group contrasts and uncertainty intervals were not disclosed in the release. Interpretation B: the biomarkers represent short-term systemic suppression without useful plaque benefit or tolerable long-term risk. The absence of imaging supports caution, while the mechanistic breadth challenges a purely nonspecific explanation. Six-month placebo-adjusted coronary imaging, biomarker-to-imaging correlations, dose response and infection data would discriminate these views.<\/p>\n<h4>Company and Product Background<\/h4>\n<p>Monte Rosa develops orally bioavailable molecular glue degraders.
